abstract

background the 30 kda major secretory protein of mycobacterium tuberculosis (antigen 85b) is a primary vaccine candidate. this secreted antigen induces a protective immune response and stimulates the production of ifn-γ in animal models. objectives the aim of this study was cloning and expression of ag 85b of m. tuberculosis in escherichia coli. materials and methods to produce recombinant ag85b, the fbpb gene was amplified by pcr method. then inserted into the pet101/d vector and transported into e. coli strain topo10. plasmid containing pet101/d: ag85b was transformed into competence e.coli bl21 (de3). the transformed e.coli strain bl21 was effectively expressed recombinant ag85b. results the expressed fusion protein was found almost entirely in the insoluble form. followed by sonication to disrupt the cells, solution of the cell debris was centrifuged and after use of ni–nta column and 6 molar urea and 6 m guanidine-hcl solutions recombinant protein was purified. conclusions these results could serve as a basis for further studies in endemic regions of tuberculosis on the usefulness of this gene and its expression product in the development of subunit vaccine and dna vaccine against tuberculosis.

Mycobacterium tuberculosis HspX/EsxS Fusion Protein: Gene Cloning, Protein Expression, and Purification in Escherichia coli

Background: The purpose of this study was to clone, express, and purify a novel multidomain fusion protein of Micobacterium tuberculosis (Mtb) in a prokaryotic system. Methods: An hspX/esxS gene construct was synthesized and ligated into a pGH plasmid, E. coli TOP10 cells were transformed, and the vector was purified. The vector containing the construct and pET-21b (+) plasmid were digested ...

Molecular Cloning, Expression and Purification of Protein TB10.4 Secreted by Mycobacterium Tuberculosis

Objective(s) Tuberculosis (TB) is the leading cause of mortality among the infectious diseases, especially in developing countries. One of the main goals in tuberculosis research is to identify antigens which have the ability of inducing cellular and/or humoral immunity in order to use them in diagnostic reagents or vaccine design. The aim of this study was to clone and express the TB'0.4 prot...

[Cloning and expression of Mycobacterium bovis secreted protein MPB51 in Escherichia coli].

The gene encoding MPB51 was amplified from M. bovis Valleel11 chromosomal DNA using PCR technique, and the PCR product was approximately 800 bp DNA segment. Using T-A cloning technique, the PCR product was cloned into pGEM-T vector and cloning plasmid pGEM-T-51 was thus constructed successfully. pGEM-T-51 and pET28a( + ) were digested by BamH I and EcoR I double enzymes. The purified MPB51 gene...

HLA-DR binding prediction and experimental evaluation of T-cell epitopes of mycolyl transferase 85B (Ag85B), a major secreted antigen of Mycobacterium tuberculosis.

OBJECTIVE To identify T-cell epitopes of Ag85B by analysis of its sequence for prediction to bind HLA-DR alleles and evaluate the predicted peptides for recognition by T cells in antigen-induced proliferation assays. MATERIALS/SUBJECTS AND METHODS The complete sequence of Ag85B was analyzed for HLA-DR binding prediction to 51 HLA-DR alleles by using a virtual matrix-based prediction program (...
